自閉癥發病機理新發現

　　4月4日，國際期刊《科學轉化醫學》（Science Translational Medicine）雜志發表了美國南加州大學的研究成果，研究者認為一段3.9kb的非編碼RNA或導致了自閉癥的發生。

　　自閉癥又稱孤獨癥，被歸類為一種由于神經系統失調導致的發育障礙，其病征包括不正常的社交能力、溝通能力、興趣和行為模式。自閉癥患者有稀有突變和拷貝數的變異，但引起這一疾病的突變尚不清楚。

　　為找到導致自閉癥的基因和信號通路，研究者做了一項GWAS調查，結果表明，自閉癥與SNP rs4307059顯著相關，rs4307059位于基因貧乏的5p14.1染色體上，5p14.1可轉錄出一段3.9kb的非編碼RNA。而這段非編碼RNA正是由膜突蛋白假基因（moesin pseudogene 1,MSNP1）的反義鏈編碼，因此研究人員將其稱為MSNP1AS（moesin pseudogene 1,antisense）。

　　MSNP1AS與膜突蛋白的RNA可互補配對，而膜突蛋白的主要功能是調節神經元結構。自閉癥患者的大腦皮層中MSNP1AS非編碼RNA的表達量是正常人的12.7倍，而自閉癥患者大腦中的膜突蛋白表達量沒有明顯改變，但在人細胞系中過量表達MSNP1AS時，膜突蛋白的表達量降低。

　　盡管自閉癥患者大腦中的膜突蛋白表達量沒有明顯改變，但自閉癥患者大腦皮層膜突蛋白表達量改變只發生在特定時期發育時期。
